Specificity:
Rabbit Anti-Human Chemokine (C-X-C Motif) Ligand 2 (CXCL2) recognizes Human CXCL2. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.

Background:
Chemokine (C-X-C motif) ligand 2 (CXCL2) is an inducible murine chemokine involved in attraction of polymorphonuclear granulocytes to sites of infection.1 This chemokine is secreted by monocytes and macrophages and is chemotactic for hematopoietic stem cells.2,3,4

Formulation
This antigen affinity purified polyclonal antibody is aseptically filtered and lyophilized from PBS with no carrier proteins or preservatives added.
Reconstitution
Aseptically reconstitute this antigen affinity purified polyclonal antibody using sterile deionized water to a concentration >0.2 mg/ml.
Storage and Stability
The lyophilized antibody should be stored desiccated at -20°C. The reconstituted antibody can be stored for at least six weeks at 4°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.5-2.0 µg/ml. A suitable detection antibody is Anti-Human CXCL2-Biotin (Antigen Aff. Pur.) (Leinco Prod. No.: G148) at a concentration of approximately 0.15-0.30 µg/ml. When used in conjunction with an optimal second step reagent such as UltraAvidin™- Horseradish Peroxidase (HRPO) (Leinco Prod. No.: A106) and a chromogenic substrate such as TMB HRPO Microwell Substrate Standard Kinetic One Component "Ready Use" (Leinco Prod. No.: T118), the sensitivity of this sandwich ELISA is 0.2-0.4 ng/well. A suggested standard for this assay is Recombinant Human CXCL3 (Leinco Prod. No.: G132).
Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5-2.0 µg/ml with a suitable second step reagent such as Goat Anti-Rabbit IgG (H&L) Horseradish Peroxidase (Leinco Prod. No.: R115). The detection sensitivity of this indirect ELISA for Human GRO-γ is approximately 0.2-0.4 ng/well.
