Description
Specificity:
Anti-MAP-2 recognizes (Mr 300 kDa) MAP-2 protein from bovine brain. The antibody strongly cross-reacts with MAP-2 from human, rat, mouse and chicken brains. Anti-MAP-2 (Clone: AP20) reacts with the high molecular weight forms (2a & 2b) of MAP-2 but not with the low molecular weight form (2c).
Antigen Distribution:
Anti-MAP-2 reacts with dendrites and cell bodies of neurons.
Background:
Microtubules are associated with a family of proteins called microtubule associated proteins (MAPs), which includes the protein τ (tau) and a group of proteins referred to as MAP1, MAP2, MAP3, MAP4 and MAP5. MAP2 is made up of two ~280kDa apparent molecular weight bands referred to as MAP2a and MAP2b. A third lower molecular weight form, usually called MAP2c, corresponds to a pair of protein bands running at ~70kDa on SDS-PAGE gels. All these MAP2 forms are derived from a single gene by alternate transcription, and all share a C-terminal sequence which includes either three or four microtubule binding peptide sequences, which are very similar to those found in the related microtubule binding protein τ (tau). MAP2 isoforms are expressed only in neuronal cells. Antibodies to MAP2 are therefore excellent markers on neuronal cells, their perikarya and neuronal dendrites. MAP2 serves to stabilize microtubules (MT) growth by crosslinking MT with intermediate filaments and other MT.
Reporter Molecule:
Fluorescein (FITC) is a small fluorescent organic molecule with a molecular weight of 473 daltons with an absorbance maximum of ~492 nm and an emission maximum of ~518 nm.

This Fluorescein (FITC) conjugate is formulated in 0.01 M phosphate buffered saline (PBS) pH 7.4, 150 mM NaCl, 1% BSA and 0.09% sodium azide as a preservative.
Storage and Stability
This Fluorescein conjugate is stable when stored at 2-8°C. Do not freeze.

IHC (NBF/Par.): This antibody should give satisfactory staining results when used at a dilution of 1:500.
Immunohistochemistry on Frozen Sections: This antibody should give satisfactory staining results when used on frozen sections at a dilution of 1:500.
